How position sensitive are skills? If an assassin is fighting a class 1v1 and it takes off in flight, is the assassin SOL? because it seems with the flying element, it'll be a lot harder to find the "back" of a person in pvp.

hmmm... flying is pretty smooth I'd say, but about your other question as in if melee could fight ranged such as sorc or ranger on the air... well we haven't tested abyss yet and people tend to duel on ground atm because of 1 min duration of flight timer. But.. well from what I've seen on some video before, gladiators or assassins didn't really have trouble beating sorc in aerial combat duel. Sure sorc can attack from few yards away but they have casting time and while they are casting it's not hard for melee classes to approach them and attack, unless you got snared or slowed by frost attack before hand.

Yes they can. Some asmodian assassin sneaked into Elyos' fortress and tried to gank a sorc then got killed last night. He later posted on forums about how he got in with out being caught, he flew over the wall while in stealth.
